The Fundamentals

As a refresher, here’s what all of the science tells us:

  • The aging process starts in our cells.
  • NAD+ is a natural compound found within every cell in our bodies. It is responsible for energy, healthy aging, and governs our Sirtuins, which are known as ‘The longevity Gene’.
  • NAD+ declines dramatically as we age and lifestyle choices alone cannot change that.
  • VO2 max, or maximal oxygen uptake, also declines as we age. But our cells need oxygen for energy and maintaining NAD+ levels. 
  • Inflammation increases as we age. Inflammation is the enemy because the body wastes precious NAD+ in the pursuit of fighting chronic inflammation.
  • Raising NAD+ levels in isolation has a counterproductive side effect. Ironically, it creates a waste product in our bodies called NAM which suppresses our Sirtuins (that all important longevity gene that we were trying to activate in the first place!).
  • Up to 90% of NAD+ that is used in the body is actually recycled. So it’s not as simple as seeking to simply synthesize new NAD+. You have to support the body’s recycling mechanism as well.

Promoting NAD+ Synthesis

Cellenial uses a superior precursor, Nicotinic Acid (NA). All NAD+ supplements use a precursor but NA is up to 3 times more effective than other precursors in raising NAD+ levels. It has an extensively researched safety profile and is natural, efficient, straightforward, reliable, and versatile.

NA is a brilliant precursor because it uses an existing pathway in the body known as the De Novo pathway to convert NA into NAD+. This pathway minimizes the effect of a Catch 22 that scientists recently discovered which goes as follows: when NAD+ is produced and consumed in the body, it can generate a waste byproduct known as NAM. NAM is a Sirtuin suppressor. If the goal of a longevity supplement is to activate our Sirtuins, then the mechanism through which NAD+ is boosted cannot be counterproductive to the entire goal of the product! Getting around this challenge is what makes Cellenial such a smart and elegant solution.

Supporting NAD+ Recycling

As a result of Cellenial using the De Novo pathway for NAD+ synthesis, capacity is freed up in the body’s NAD+ recycling pathways. Given the fact that up to 90% of the NAD+ used by your body is actually recycled, this is essential. You could say that supporting effective recycling is just as important as the synthesis component.

Our precursor, NA, also has a “flushing” effect which improves circulation. This distributes more oxygen-rich blood throughout the body. Because oxygen fuels the body’s recycling mechanism for NAD+, better circulation means better NAD+ metabolism.

R-Lipoic Acid

R-Lipoic acid, the biologically active form of alpha-lipoic acid, has demonstrated anti-inflammatory properties through several mechanisms. Its anti-oxidant activity can neutralize free radicals and reduce oxidative stress, which is a significant contributor to the inflammatory process. By decreasing oxidative stress, R-Lipoic acid helps mitigate the damage to cells and tissues that can trigger inflammatory responses.
NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) is a protein complex that plays a crucial role in regulating the immune response to infection. Overactivation of NF-κB leads to chronic inflammation. R-Lipoic acid has been shown to inhibit the activation of NF-κB, thereby reducing the expression of pro-inflammatory cytokines such as TNF-α (tumor necrosis factor alpha) and IL-6 (interleukin-6).

Keeping inflammation down means a lot of NAD+ is preserved, making more available for other purposes.

L-Citrulline and Alpha Ketoglutarate (AKG)

L-Citrulline exhibits anti-inflammatory properties by suppressing the NF-κB pathway, reducing the expression of inflammatory cytokines that promote chronic inflammation. It also improves blood flow through its vasodilatory effects, allowing for increased circulation to areas of the body that often don’t have enough flow. 

AKG also has anti-inflammatory effects by suppressing the NF-κB pathway. As part of the Citric Acid cycle it also plays a role in creating ATP, another key energy molecule your body needs.

Together, these two are highly effective in further reducing chronic inflammation, protecting NAD+ and supporting ATP production.

Berberine

Berberine can directly activate SIRT1, one of the most important of the Sirtuins genes. It can indirectly enhance NAD+ availability and utilization through its effects on SIRT1, AMPK, and its antioxidant and anti-inflammatory activities. It even enhances mitochondrial function and stimulates mitochondrial biogenesis.

Recently, Berberine has been shown to reduce Arterial plaque. Research here is ongoing, but we are very excited about the early results we’re seeing with Berberine. Berberine activates AMP-activated protein kinase (AMPK), an enzyme that plays a key role in regulating cellular energy homeostasis. AMPK activation helps to balance energy production and consumption within cells, enhancing mitochondrial efficiency and protecting cells from energy-related stress. Berberine can help lower blood sugar levels in people with type 2 diabetes by improving insulin sensitivity and increasing glucose uptake in cells.

Acetyl L-Carnitine

Carnitine is an important molecules that transports fatty acids into your mitochondria, providing them with more fuel to make ATP. ALC readily crosses the blood-brain barrier and has antioxidant and anti-inflammatory effects. It can improve mental and physical fatigue in older adults, and we can all certainly use a little more energy.

Garlic Extract

Improves blood flow primarily due to its allicin content, which is used to make nitric oxide that promotes vasodilation. Garlic has been shown to reduce platelet aggregation and improve endothelial (blood cell) function in general.
